Zan is one of the 3 Sanada demons, extremely powerful ninjas, who act under the guidance of Misanagi, the Okashira (leader) of the Sanada. He fights alongside his comrades (the other two demons), named Ren and Baku, when Himura Kenshin and his friends, assisted by a former Sanada member, named Kaita, attempt to thwart the plans of Misanagi. He encounters Sagara Sanosuke and on 2 occassions, he nearly kills Sano, but the latter is saved by Tsukayama Yutarō and Kamiya Kaoru in both instances. Sano is able to take advantage of the instant when Kaoru distracts Zan and at that moment he attacks using Futae no Kiwami with his right elbow (due to the fact that the move has seriously damaged his right hand over-time).
